Can a owner/builder be issued a permit to construct a two-family threshold building?

No, 110.8.5 is a specific requirement and does not include owner builders.
Commentary
In addition to the Building Code, the basis for this requirement originates in Florida Statute 553.79 (8) which prohibits issuance of a permit except to a CGC, or, in some cases, a CBC
(8) No enforcing agency may issue a building permit for construction of any threshold building except to a licensed general contractor, as defined in s. 489.105(3)(a), or to a licensed building contractor, as defined in s. 489.105(3)(b), within the scope of her or his license.
