Is 606.2 to be interpreted to require a separate water shut-off valve in addition to providing a water stop at the fixture location?

No. Section 606.2 requires a shutoff valve at the fixture supply to each plumbing fixture, with a few exceptions for specific occupancies. If the shutoff valve is provided on the supply side of the fixture in the form of a water stop, or angle stop, the code has been met. A second valve or supplemental shut off valve is not required.
Commentary
This question seems to come up often (see BOAF Interpretations 8687 and 8808.) It was also the subject of a Declaratory Statement DS 2025-003, earlier this year.
